Seminar Title:Genome-Wide Transposon Mutagenesis Identifies Genes That Transform Neural Stem Cells to Cancer-Initiating Cells

Summary: Genome-wide insertional mutagenesis is an unbiased and high-throughput method for the cancer gene discovery. Neural stem cells (NSCs) are considered as the cell-of-origin of glioma-initiating cells. To identify genetic pathways responsible for the transformation of NSCs to cancer-initiating cells&s_comma; we used Sleeping Beauty transposon-based insertional mutagenesis. We show that transposons facilitate immortalization of NSCs as well as tumor formation. Analysis of common insertion sites in 67 tumors identified 149 gene loci&s_comma; which include brain tumor-associated genes such as Met and Nf1&s_comma; as well as candidate driver genes. Comparisons of insertions between transplanted cells and tumors revealed that a small number of cells that acquired transposon insertions in Met&s_comma; etc. clonally expanded to form tumor. We show that transposon-based insertional mutagenesis is a powerful tool for functional genomics of cancer.
