The synthesis of messenger RNA by RNA polymerase II is
controlled by the interplay of a large number of transcriptional
regulatory proteins&s_comma; including DNA binding transcription factors&s_comma; the
basal initiation factors&s_comma; and various coregulatory complexes that
function at both initiation and elongation. Some coregulatory
complexes&s_comma; such as the Mediator of RNA polymerase II transcription&s_comma;
contribute to transcriptional regulation by transmitting signals from
DNA binding transcription factors to RNA polymerase II and the
general initiation factors. Others&s_comma; including chromatin modifying
and remodeling complexes&s_comma; function by altering chromatin structure to
generate localized chromatin environments that are more or less
conducive to transcription. We have used a MudPIT-based proteomics
approach to define a family of chromatin modifying and remodeling
complexes that each include a SNF2-like helicase&s_comma; the actin-related
protein Baf53/Arp4&s_comma; and the AAA+ ATPases Tip49a and Tip49b. This
talk will focus on studies of the human INO80 chromatin remodeling
complex&s_comma; a member of this family. I will describe experiments
demonstrating a role for the INO80 complex as an essential
coregulator for the transcription factor YY1&s_comma; as well as experiments
exploring the contribution of an INO80-associated deubiquitinating
enzyme in INO80 function.
