 Recent developments suggest that epigenetic mechanisms may explain a
number of epidemiological and clinical findings in schizophrenia (SCZ).
The epigenetic research program indicates that gene activity is
critically important for normal functioning of the genome&s_comma; and
dysregulation of such gene activity may increase the risk to be affected
with SCZ. Epigenetics provides a new interpretation for a series of
unclear issues in SCZ such as the discordance of monozygotic twins&s_comma;
sexual dimorphism&s_comma; fluctuating course&s_comma; and cases of recovery&s_comma; among
others. In addition&s_comma; numerous molecular aberrations identified in SCZ
are consistent with putative epigenetic defects. Examples for this
statement could be dopamine D2 and serotonin 2A receptor genes&s_comma; which
have been subjected to detailed pharmacological&s_comma; imaging&s_comma; and genetic
studies&s_comma; however&s_comma; it remains unclear what molecular mechanisms cause the
changes in density of the D2 and HT2A receptors. Apart from the
candidate genes&s_comma; epigenetic strategies are relevant to understanding the
role of the non-coding part of genome in SCZ. This applies to unstable
trinucleotide repeats and retroelements. It is becoming clear that
pathological effects of unstable trinucleotide repeats as well as
endogenous retroelements significantly depend on their epigenetic
status&s_comma; which has never been taken into account in SCZ studies.
Epigenetic strategies are new developments in SCZ studies that may be of
major value in the light of slow progress in traditional linkage and
association strategies of SCZ thus far.
