演題: LAR family PTPases: association with liprins and a rho exchange factor

要旨:LAR family transmembrane PTPases function in axon guidance and mammary gland development. To understand how these PTPases function&s_comma; we screened for LAR binding proteins. One of the LAR binding proteins isolated&s_comma; liprin-alpha1 colocalizes with LAR at discrete ends of focal adhesions&s_comma; implicating the LAR/liprin-alpha1 complex in the regulation of cell-matrix interactions.

 Liprin-alpha1 is the prototype member of a family of LAR binding proteins that may function to localize LAR family PTPases at specific sites on the plasma membrane&s_comma; possibly regulating their interaction with the extracellular environment and their association with substrates. The second LAR binding protein isolated&s_comma; Trio&s_comma; is a complex protein possessing two rho/rac guanine nucleotide exchange factor (GEF) domains. NIH 3T3 cells stably expressing the individual GEF domains exhibit distinct cell morphologies. Furthermore&s_comma; cells expressing the N-terminal GEF domain display more rapid cell spreading&s_comma; haptotactic cell migration&s_comma; and anchorage-independent growth&s_comma; indicating that Trio regulates both cell motility and cell growth.

Thus&s_comma; Trio appears to integrate and amplify signals involved in coordinating actin remodeling and focal contact formation necessary for cell migration and growth.

Possibly&s_comma; a LAR/Trio complex coordinates actin reorganization and membrane trafficking that occurs at the leading edge of motile cells and neural growth cones.
